What is Tukysa?

Tukysa is an oral prescription medication used to treat a specific type of breast cancer. It is designed for adults with HER2-positive breast cancer that is either locally advanced or has spread to other parts of the body (metastatic). Tukysa contains the active ingredient tucatinib, which is a type of drug known as a kinase inhibitor. This medication is not used by itself. It is always given in combination with two other cancer drugs: trastuzumab and capecitabine. This three-drug combination provides a powerful approach to managing advanced breast cancer. It is typically prescribed after a patient has already tried at least one other anti-HER2 drug regimen.

How does Tukysa work?

Tukysa works by targeting and blocking a protein called human epidermal growth factor receptor 2 (HER2). In HER2-positive breast cancer, cancer cells have too much of this protein on their surface. The HER2 protein acts like an accelerator, sending signals that tell the cancer cells to grow and divide uncontrollably. Tukysa gets inside the cancer cell and specifically blocks the kinase function of the HER2 protein. This action stops the growth signals. By blocking these signals, Tukysa helps to slow or stop the growth and spread of cancer cells. It works together with trastuzumab, which targets HER2 from outside the cell, and capecitabine, a chemotherapy that kills cancer cells.

Who is Tukysa for?

Tukysa is intended for adult patients diagnosed with HER2-positive breast cancer. It is specifically for cases where the cancer is either advanced and cannot be surgically removed or has metastasized (spread) to other areas of the body, including the brain. This medication is an option for patients whose cancer has continued to grow after receiving at least one previous anti-HER2-based treatment regimen. It is suitable for both men and women. Before starting treatment, a doctor must confirm that the cancer is HER2-positive through laboratory testing of the tumor tissue. Patients must be able to tolerate combination therapy with trastuzumab and capecitabine.

Dosage and administration

Tukysa dosage and administration must be directed by a qualified healthcare provider. The standard dose is taken orally twice a day, approximately 12 hours apart. Patients should swallow the tablets whole and should not chew, crush, or split them. Tukysa can be taken with or without food. Consistency is important, so you should take it at the same times each day. If you vomit after taking a dose, do not take an extra dose. Simply wait until your next scheduled time. Tukysa is part of a combination treatment. It is taken along with trastuzumab, which is given by infusion, and capecitabine, another oral medication.
