Plenvu (generic name: macrogol 3350) is a laxative specifically formulated to cleanse the bowel prior to procedures like colonoscopies. It combines macrogol 3350 (also known as polyethylene glycol 3350) with electrolytes such as sodium ascorbate, sodium sulfate anhydrous, ascorbic acid, sodium chloride, and potassium chloride. This combination works by drawing water into the bowel, resulting in watery diarrhea that effectively clears the intestinal tract, ensuring a clear view during medical examinations. Belonging to the class of osmotic laxatives, Plenvu operates by osmosis—drawing water into the colon to soften stools and stimulate bowel movements. The inclusion of electrolytes helps maintain the body’s electrolyte balance during the cleansing process, reducing the risk of dehydration. Plenvu is intended for adults aged 18 and over who require bowel cleansing before clinical procedures requiring a clear bowel. Proper bowel preparation is crucial for the accuracy of these procedures, allowing for the detection of abnormalities such as polyps or lesions.

Usual Dosage(s) and Mode(s) of Administration

Plenvu is supplied as a powder that must be reconstituted with water before ingestion. Read and follow the instructions for preparation and dosing. The treatment consists of two separate doses, which can be administered using different dosing schedules as directed by the prescribing physician:
  • Two-Day Split-Dosing Regimen: The first dose is taken in the evening before the procedure (between 4 pm and 8 pm), and the second dose is taken the next morning, approximately 12 hours after the first dose.
  • One-Day Morning Dosing Regimen: Both doses are taken on the morning of the procedure, with at least a 2-hour interval between the start of the first and second doses.
To prepare each dose:
  1. Empty the contents of the dose into the provided mixing container.
  2. Add water up to the fill line (approximately 16 fluid ounces).
  3. Stir or shake the solution until completely dissolved (this may take 2 to 3 minutes).
  4. Drink the solution over 30 minutes.
  5. After finishing the solution, refill the container with clear liquids up to the fill line and drink over the next 30 minutes.
It’s important to consume additional clear liquids during the process to prevent dehydration and ensure effective bowel cleansing.

Storage Guidelines

Before reconstitution, Plenvu sachets should be stored below 25°C (77°F) in a dry place, away from direct sunlight. After dissolving the powder in water, the solution may be stored at room temperature (below 25°C) or refrigerated (2°C to 8°C). However, it should be consumed within 24 hours of preparation to ensure effectiveness and safety. If in any doubt, consult our blog on safe medicine management.

Common Questions About Plenvu

Are there any dietary restrictions while using Plenvu?
Yes, dietary modifications are essential for effective bowel preparation with Plenvu. Patients are usually advised to consume a light breakfast followed by a light lunch on the day before the procedure. After lunch, only clear fluids should be consumed until after the colonoscopy. Clear fluids include water, clear soups, herbal tea, black tea or coffee (without milk), and clear fruit juices (without pulp). Solid foods and colored beverages, especially those that are red or purple, should be avoided during this period.
Is macrogol 3350 safe for long-term use?
Macrogol 3350 is generally considered safe for short-term use, particularly for bowel preparation before medical procedures. For chronic constipation, its long-term use should be under medical supervision to prevent potential electrolyte imbalances or dependence. It’s important to consult a healthcare provider for personalized advice regarding prolonged use.
What happens if the bowel isn’t properly cleansed before a colonoscopy?
If the bowel isn’t adequately cleansed, residual stool can obscure the colon’s lining, making it challenging to identify abnormalities. This may result in incomplete examinations, missed lesions, or polyps, and could necessitate rescheduling the procedure. Proper bowel preparation is essential for a thorough and accurate colonoscopy.

  • Can macrogol 3350 cause dehydration?
As an osmotic laxative, macrogol 3350 draws water into the bowel to soften stools, which can lead to increased water loss through bowel movements. To mitigate the risk of dehydration, it's essential to maintain adequate fluid intake while using this medication. Healthcare providers often recommend consuming clear fluids during the bowel preparation process to ensure proper hydration.
  • Why is bowel preparation necessary before a colonoscopy?
Bowel preparation is crucial before a colonoscopy to clear the colon of any stool, allowing for accurate visualization of the intestinal lining. This clarity enables healthcare providers to detect abnormalities such as polyps, lesions, or signs of colorectal diseases. Inadequate bowel preparation can obscure the view, potentially leading to missed diagnoses or the need to repeat the procedure.
  • What are osmotic laxatives, and how do they differ from other laxatives?
Osmotic laxatives, such as macrogol 3350, work by drawing water into the bowel, softening the stool, and promoting bowel movements. This mechanism is different from stimulant laxatives, which stimulate the intestinal muscles to contract, and bulk-forming laxatives, which increase stool bulk by absorbing water. Osmotic laxatives are generally considered gentle and effective for relieving constipation.
  • How long does it take for osmotic laxatives to work?
Osmotic laxatives typically start to work within 2 to 3 days after ingestion. However, the onset of action can vary depending on the specific product and individual response. It's important to follow the dosing instructions provided by the healthcare provider or as indicated on the product packaging
